Background
There are 8 site (9nt) differences between W3110 and MG1655. They reside in 7 orgs and one rRNA gene. Two are nonfunctional (rpoS and dcuA) and 5 are unknown missense mutations. bcsQ allele (codon 6 changed from TAG (stop) to TTG (leucine)).
This strain is used for biofilm formation experiments and is capable of expressing cellulose and curli at temperatures of less than 30°C in LB without salt.
Paper that describes the generation of the strain.
Cultivation and freezing protocols
Strain should ideally be stored in 7% DMSO (930uL overnight culture and 70uL DMSO in cryotube), alternatively can be stored in glycerol stock (25% v/v) (500uL overnight culture and 500uL 50% glycerol), flash-frozen in liquid nitrogen. Long-term storage -80 degrees C and DMSO storage recommended.
