Type of measurement:
Liquid chromatography for protein purification/analysis in aqueous solutions at medium pressure (< 5 bar = 0.5 MPa) with automated fraction collector
- Application depends on column used (e.g. size exclusion, affinity, ion exchange, etc.)
- Detectors: UV (190-700 nm), conductivity
- Sample volumes: 5 µL - 5 mL (sample loop) up to several liters (pump inlet A3)
- Flow rates: 0.001 - 25 mL/min
Requirements:
- Gain permission to work in the Biolabs by follwoing the How-to-Start procedure.
- Get access to the BCP-storage network drive suprafab-analytics to save measuremnt data.
- Book the device using OpenIRIS (protocls see here).
- Get an introduction to the device by the responsible person (see above).
- All buffers and solutions must be prepared with MilliQ, filtered (0,22µm) and degassed before use (simultaneous filtering and degassing can be done in E030)
- Read up on the properties of your column matrix (capacity, pore size, chemical resistance, max pressure, etc)
- Get familiar with the flow path
